TLR2 in γδ T cells is required for metabolic support of their IL-17A response to a commensal bacterium on the ocular surface

چکیده
Abstract The ocular surface is colonized by commensal microbiota, which tune the local mucosal immune response. We previously identified an commensal, Corynebacterium mastitidis (C. mast), induced IL-17A production from γδ T cells in conjunctiva and eye-draining lymph nodes (DLN), enhanced host resistance to fungal bacterial pathogens. To elucidate how producing (γδT17) are activated C. mast, we associated WT TLR2−/− mice with investigated role of TLR2 this process. This was based on rationale that mastis a Gram-positive bacterium expression γδT increased after mast association. Mechanistic studies showed required not only DCs (for IL-1 production) but also cells. RNA-Seq revealed genes related mitochondrial oxidative phosphorylation were dysregulated γδT17 cells, indicating engagement may regulate metabolic processes Seahorse assay confirmed T17 displayed impaired oxygen consumption rates lower ATP production. Th17 cells are not required for maintenance of IL-17A producing γδ T cells in vivo
γδ T cells producing interleukin-17A (γδT17) are thought to develop spontaneously in the thymus and to be maintained in the periphery. Previous studies suggested a role for T-helper 17 (Th17) cells in the maintenance of γδT17 via the expression of transforming growth factor-β1 (TGFβ1). However, we have previously found that Th17 cells were not required for expansion of γδT17 cells after lung tr...
متن کاملCD3bright signals on γδ T cells identify IL-17A-producing Vγ6Vδ1+ T cells.
Interleukin-17A (IL-17A) is a pro-inflammatory cytokine that has an important role at mucosal sites in a wide range of immune responses including infection, allergy and auto-immunity. γδ T cells are recognized as IL-17 producers, but based on the level of CD3 expression, we now define the remarkable ability of a CD3(bright) γδ T-cell subset with an effector memory phenotype to rapidly produce I...
متن کاملγδ T Cells Are Required for Pulmonary IL-17A Expression after Ozone Exposure in Mice: Role of TNFα
Ozone is an air pollutant that causes pulmonary symptoms. In mice, ozone exposure causes pulmonary injury and increases bronchoalveolar lavage macrophages and neutrophils. We have shown that IL-17A is important in the recruitment of neutrophils after subacute ozone exposure (0.3 ppm for 24-72 h). We hypothesized that γδ T cells are the main producers of IL-17A after subacute ozone. To explore t...
متن کاملIL-17A-producing resident memory γδ T cells orchestrate the innate immune response to secondary oral Listeria monocytogenes infection.
Memory γδ T cells are important for the clearance of Listeria monocytogenes infection in the intestinal mucosa. However, the mechanisms by which memory γδ T cells provide protection against secondary oral infection are poorly understood. Here we used a recombinant strain of L. monocytogenes that efficiently invades the intestinal epithelium to show that Vγ4(+) memory γδ T cells represent a resi...
متن کاملMicroglia Induce Neurotoxic IL-17+ γδ T Cells Dependent on TLR2, TLR4, and TLR9 Activation
BACKGROUND Interleukin-17 (IL-17) acts as a key regulator in central nervous system (CNS) inflammation. γδ T cells are an important innate source of IL-17. Both IL-17+ γδ T cells and microglia, the major resident immune cells of the brain, are involved in various CNS disorders such as multiple sclerosis and stroke. Also, activation of Toll-like receptor (TLR) signaling pathways contributes to C...
